"Medicinal plants, called "Sarang Semut", myrmecodia pendens Merr. & Perry (Rubiaceae) is an epiphyt traditionally used in Papua for healing various diseases such as hyperuricemia, reumathic, and cancer. A chemical compound recognized as xanthinoxidase inhibitor has been isolated from n-buthanol fraction of the palnt extrac. The isolation was carried out by column chromatography (Sio,CHCL - MeOH = 5 : 1-1 : ) and the purification was conducted using high presure liquid chromatography (RP,C - 18 ; MeOH - = 5 : 1)). The activity test for xanthinoxidase inhibitor was performmed by using modified Noro method. The pure compound showed xanthinoxidase inhibitor activity of IC at 79,77%"

"Xantin oksidase mengkatalisis oksidasi hipoxantin dan xantin menjadi asam urat. Kadar asam urat yang meningkat memiliki keterkaitan terhadap penyakit gout. Gout merupakan kelainan metabolik pada katabolisme purin. Salah satu tumbuhan obat yang telah dilaporkan mempunyai efek terhadap penyakit gout dan sebagai anti inflamasi adalah tanaman Sidaguri (Sida rhombifolia L.).
Tujuan penelitian ini untuk menguji kemampuan daun Sidaguri dalam menghambat aktivitas xantin oksidase dan identifikasi golongan kandungan kimianya. Serbuk simplisia diekstraksi berturut-turut dengan cara maserasi bertingkat menggunakan empat pelarut berdasarkan tingkat kepolaran, yaitu petroleum eter, etil asetat, n-butanol, dan etanol 96%. Pengujian penghambatan aktivitas xantin oksidase dilakukan dengan metode spektrofotometri.
Berdasarkan uji penghambatan aktivitas xantin oksidase, semua ekstrak dapat menghambat aktivitas xantin oksidase dengan nilai IC50 1,71 μg/mL pada ekstrak n-butanol; IC50 2,38 μg/mL pada ekstrak etil asetat; IC50 4,64 μg/mL pada ekstrak etanol; dan IC50 9,52 μg/mL pada ekstrak petroleum eter. Pada plot Lineweaver-Burk menunjukkan jenis penghambatan enzim pada ekstrak n-butanol adalah kompetitif. Hasil uji identifikasi kimia pada ekstrak daun sidaguri menunjukkan adanya alkaloid, glikosida, flavonoida, dan terpen.

The xanthine oxidase catalyses the oxidation of hypoxanthine to xanthine and then to uric acid. Increasing uric acid levels have related to gout. Gout is a metabolic disorder in the catabolism of purines. One of the herbs that has been reported to have effect on gout and as an anti-inflammatory is sidaguri (Sida rhombifolia L.).
The purpose of this study was to test the ability of leaf Sidaguri in inhibiting xanthine oxidase activity and identification of chemical constituents. The sample was macerated respectively with petroleum ether, ethyl acetate, nbuthanol, and 96% ethanol. Inhibition of xanthine oxidase activity test carried out by spectrophotometric methods.
Based on xanthine oxidase inhibitory activity test, all the plant extracts were active in inhibiting xanthine oxidase with IC50 value of 1,71 μg/mL on n-buthanol extract; IC50 value of 2,38 μg/mL on ethil acetat; IC50 value of 4,64 μg/mL on 96% ethanol; and IC50 value of 9,52 μg/mL on petroleum ether extract. The Lineweaver-Burk plots showed that the type of nbuthanol extract was a competitive inhibition. The results of chemical identification on a sidaguri leaf extract contain alkaloids, glycoside, flavonoids, and terpenes."
