"Dengue merupakan infeksi virus yang sering ditemukan pada daerah tropis seperti Indonesia. Infeksi ini adalah sebuah masalah kesehatan dengan lebih dari 200 juta orang berisiko terinfeksi dan diperkirakan 30 juta orang terinfeksi dengue setiap tahunnya. Dengan case fatality rate 2,5%, 2000 orang di Indonesia meninggal setiap tahunnya karena infeksi dengue. Sampai saat ini, belum ada pengobatan khusus untuk dengue yang dapat mengurangi aktivitas atau jumlah virus seperti antivirus. Perkembangan antivirus dengue terbilang lambat, dibandingkan dengan virus flaviviridae lain, seperti hepatitis C. Untuk mengatasi masalah ini, peneliti mencoba mencari pengobatan yang mungkin untuk menjadi antivirus dengue dengan daun trembesi (Samanea Saman). Percobaan dilakukan menggunakan sel Huh7it-1 yang diinfeksikan virus dengue lalu diberikan konsentrasi ekstrak yang beragam. Keamanan ekstrak yang dilihat dari nilai CC50 didapatkan dengan metode MTT assay. Dihitung juga tingkat inhibisi IC50 yang didapatkan dengan metode Focus Assay. Hasilnya adalah CC50 201,21 dengan IC50 14,6 dan didapatkan Selectivity Index sebesar 13,7. Studi ini memperlihatkan kemampuan inhibisi replikasi dengue pada ekstrak daun trembesi dan dapat menjadi pertimbangan untuk pengembangan antivirus dengue.
Dengue is a virus infection that is usually found in tropical area such as Indonesia. With over 200 million people at risk and 30 million people infected every year, dengue is a health problem. With 2,5% case fatality rate, over 2000 people in Indonesia die everyday because of dengue infection. Until now, there has been no specialized treatment for dengue that can lower the activity or virus titer like antivirus. If compared with other flaviviridae, dengue antivirus development is slower. Because of that, researcher is seeking for antivirus through Raintree leaves extract (Samanea Saman). The experiment is using infected Huh7it-1 cells and the Cells is given varied doses of the extract. Safety of the extract is evaluated in CC50 level through MTT Assay Method. IC50 is evaluated through Focus Assay. The results are CC50 201,21ug/ml and IC50 14,6ug/ml. Therefore; the selectivity index is 13,7. This study shows dengue replication inhibition activity of Samanea Saman leaves extract, thus can be beneficial for dengue antivirus development."
