"Asam vanilat atau asam 4-hidroksi-3-metoksi benzoat merupakan derivat senyawa fenolat terhidroksilasi yang merupakan bentuk teroksidasi dari vanilin. Senyawa asam vanilat biasa digunakan sebagai agen penyedap rasa yang dilaporkan memiliki aktivitas sebagai antioksidan. Namun aktivitas antioksidan tersebut masih kurang kuat apabila dibandingkan dengan senyawa antioksidan standar, sehingga diperlukan modifikasi struktur. Sehingga pada penelitian ini dilakukan modifikasi struktur melalui reaksi Mannich atau aminoalkilasi dengan menggunakan senyawa amin N-metilpiperazin. Tujuan dari sintesis ini adalah untuk memperoleh senyawa baru sebagai alternatif antioksidan.
Sintesis dilakukan melalui 2 tahap, yaitu pembuatan asam vanilat dari vanilin, dan kemudian asam vanilat yang diperoleh disubstitusi dengan basa Mannich N-metilpiperazin.Senyawa asam vanilat yang dihasilkan merupakan serbuk putih kecoklatan dengan rendemen 91,41. Sedangkan hasil akhir reaksi tahap 2 merupakan kristal putih kecoklatan berbentuk jarum dengan rendemen sebesar 36,58.
Kemurnian dari senyawa ini diuji dengan menggunakan kromatografi lapis tipis dan titik lebur. Kemudian elusidasi senyawa dilakukan dengan spektrofotometri FT-IR, spektrometri 1H-NMR dan spektrometri 13C-NMR. Hasil uji aktivitas antioksidan metode DPPH memberikan hasil IC50 asam vanilat 191,954 M, IC50 AV-MP 14,806 M., dan senyawa standar kuersetin 11,950 M.
Vanillic acid or 4 hydroxy 3 methoxy benzoic acid is a derivatives of hydroxylated phenolic compunds which is an oxidized form of vanilllin. Vanillic acid compounds commonly used as flavoring agent which is also reported to also act as an antioxidant. However, that antioxidant activity is still less than optimal when compared to standard antioxidant compounds. Therefore structural modification of vanilic acid is required. Structural modification which can be done through Mannich reaction or alminoalkylation using amin N metilpiperazin. An object of this synthesis research was to obtain new discovered compound as alternative antioxidant. The researched method was divided by 2 phase. The first phase was to manufacture vanilat acid from vanillin, and then vanillic acid obtained was substituted with 1 methyl piperazine compound. Vanillic acid compound produced with this method is a brownish white powder with rendement of 91,41. And the final result of phase 2 reaction is a needle shaped brownish white crystalline with rendement of 36,58. The purity of this compound tested by using thin layer chromatography and melting point. Then elucidation of the compound performed with FT IR spectrophotometry, 1H NMR spectrometry and 13C NMR spectrometry. Antioxidant activity result by DPPH method is IC50 vanilic acid 191,954 M , IC50 AV MP 14,806 M., and standard compound quersetin 11,950 M."
