"This study was performed to evaluate the development of somatic embryo from embryogenic calli of ginger meristem culture. Completely randomized design was applied, replicated 4 times. Embryogenic calli from meristem tissue of inner shoot bud of rhizome obtained on MS medium containing 100 mg/L glutamine, 2% sucrose with the addition of 1.0 mg/L 2,4-D and 3.0 mg/L BA, were subjected to proliferation medium, MS and N basal media containing 3% mannitol. Then, transferred into somatic embryo maturation medium, either MS or N basal media supplemented with 6% sucrose. The number of somatic embryos-formed significantly affected by the proliferation medium applied. The highest number of somatic embryos (about 82.0 per 1 g friable calli) was achieved on the MS medium, 4 weeks after incubation. In addition, the optimum growth of embryogenic calli containing somatic embryos was obtained on MS and N medium supplemented with 6% sucrose. There were significantly difference between the media applied (MS and N ) to somatic embryos maturation. The highest number of mature somatic embryos (57.2 embrios) was achieved on the MS medium, 18 days after incubation."

"Latar belakang: Keluhan-keluhan fisik atau somatik yang mengganggu, yang tidak dapat dijelaskan secara medis, sering terjadi di masyarakat, termasuk juga kelompok mahasiswa. Mahasiswa kedokteran memiliki prevalensi gangguan psikosomatis yang lebih tinggi jika dibandingkan dengan populasi umum atau kelompok mahasiswa pada bidang akademis lainnya. Masalah kesehatan jiwa yang bermanifestasi dalam keluhan fisik dapat mengganggu kualitas hidup seorang mahasiswa. Penelitian ini menilai korelasi antara resiliensi dan gejala somatisasi pada mahasiswa kedokteran.
Metode: Desain yang digunakan pada penelitian ini adalah potong lintang. Subjek penelitian adalah mahasiswa Fakultas Kedokteran Universitas Indonesia tingkat 3. Resiliensi dan gejala somatisasi pada subjek penelitian didapatkan melalui pengisian kuesioner secara daring. Penilaian resiliensi dilakukan dengan kuesioner CD-RISC. Sedangkan, penilaian gejala somatisasi dilakukan dengan kuesioner SCL-90. Data resiliensi dan gejala somatisasi dianalisis korelasinya menggunakan Uji Spearman.
Hasil: 116 responden dari mahasiswa Fakultas Kedokteran Universitas Indonesia tingkat 3 telah mengisi kuesioner secara daring. Dengan uji Kolmogorov-Smirnov, diketahui bahwa sebaran data resiliensi normal (p>0,05). Skor rata-rata dari resiliensi responden adalah 69,39 ± 14,11. Sedangkan, skor median dari gejala somatisasi adalah 3,5 (0 – 48). Didapatkan sebaran data gejala somatisasi tidak normal (p<0,05) dengan uji Kolmogorov-Smirnov. Uji korelasi Spearman menunjukkan adanya korelasi signifikan negatif secara lemah antara resiliensi dan gejala somatisasi (p<0,05 dan r= -0,371).
Kesimpulan: Terdapat korelasi signifikan negatif secara lemah antara resiliensi dan gejala somatisasi pada mahasiswa Fakultas Kedokteran Universitas Indonesia tingkat 3. Semakin tinggi resiliensi, maka semakin rendah gejala somatisasi yang dialami.

Introduction: Disturbing physical or somatic complaints, which cannot be explained medically, often occur in the community, including college students. Medical students have a higher prevalence of psychosomatic disorders when compared to the general population or college students in other academic fields. Mental health problems that manifest in physical complaints can interfere with the quality of life of a student. This study assesses the correlation between resilience and somatic symptoms in medical students.
Methods: The design used in this study is cross-sectional. The research subjects are third level students of the Faculty of Medicine University of Indonesia. Resilience and somatic symptoms in the research subjects were obtained through filling out online questionnaires. Resilience assessment was carried out using the CD-RISC questionnaire. Meanwhile, the assessment of somatic symptoms was carried out using the SCL-90 questionnaire. Data on resilience and somatization symptoms were analyzed for correlation using Spearman's test.
Results: 116 respondents from third level of students of the Faculty of Medicine University of Indonesia have filled out online questionnaires. With the Kolmogorov-Smirnov test, it is known that the distribution of resilience data is normal (p>0.05). The average score of the respondents' resilience is 69.39 ± 14.11. Meanwhile, the median score of somatic symptoms is 3.5 (0 – 48). The data distribution of somatic symptoms is not normal (p<0.05) using the Kolmogorov-Smirnov test. The Spearman correlation test showed a weakly significant negative correlation between resilience and somatization symptoms (p<0.05 and r= -0.371).
Conclusion: There is a weakly significant negative correlation between resilience and somatic symptoms in third level students of the Faculty of Medicine University of Indonesia. The higher the resilience, the lower the somatic symptoms experienced.

"Circulating tumor cell (CTC) merupakan intermediet proses metastasis kanker yang dapat dimanfaatkan untuk diagnosis, prognosis, dan target pengobatan kanker. Pengembangan pemanfaatan CTC dapat dilakukan dengan penelitian yang umumnya melibatkan proses kultur. Medium bebas serum dinilai lebih baik dibanding medium berserum karena dapat menghasilkan data yang lebih konsisten, sehingga lebih cocok digunakan untuk penelitian yang mengkaji aktivitas fisiologi sel dan persinyalan molekular. Namun, medium bebas serum memerlukan suplemen agar sel dapat tumbuh optimum. Penambahan suplemen insulin-transferrin-selenium (ITS) telah diketahui memiliki peran penting dalam kultur sel keratosit, ovarium, dan keratinosit. Namun, belum diketahui peran ITS dalam medium bebas serum untuk kultur CTC. Penelitian ini mengkaji perbedaan efek FBS dan ITS dengan konsentrasi 1X dan 10X dalam medium bebas serum terhadap CTC yang diisolasi dengan metode eritrolisis. Kultur dilakukan selama 18 hari. Dinamika CTC dan leukosit diamati dengan meninjau viabilitasnya pada 6 hari pertama kutur. Selain itu, observasi morfologi dilakukan seiring dengan pengukuran morfometri sel. Pada hari ke-18, keberadaan CTC diverifikasi dengan imunofluoresens menggunakan marka cytokeratin 20 (CK20) dan plastin 3 (PLS3). Hasil penelitian menunjukkan bahwa CTC yang dikultur pada medium dengan penambahan 10X ITS memiliki diameter sel yang lebih besar dari yang dikultur pada medium dengan penambahan 1X ITS dan 10% FBS. Hal tersebut menunjukkan bahwa ITS memiliki peran penting dalam kultur CTC dalam medium bebas serum dan dalam konsentrasi 10X dapat meningkatkan pertumbuhan CTC kanker kolorektal.

Circulating tumor cells (CTCs) are intermediates in the cancer metastasis process and hold potential for use in cancer diagnosis, prognosis, and treatment targeting. The development of CTC applications typically involves research incorporating cell culture processes. In cell culture, serum-free media are considered superior to serum-containing media as they yield more consistent data, making them more suitable for studies examining cell physiological activity and molecular signaling. However, serum-free media require supplementation to ensure optimal cell growth. The addition of insulin-transferrin-selenium (ITS) supplements is known to play a crucial role in the culture of keratocytes, ovarian cells, and keratinocytes. However, the role of ITS in serum-free media for CTC culture remains unknown. This study investigates the differential effects of fetal bovine serum (FBS) and ITS at concentrations of 1X and 10X in serum-free media on CTCs isolated via erythrolysis. Cultures were maintained for 18 days, with CTC and leukocyte dynamics observed by assessing cell viability during the first six days of culture. Additionally, morphological observations and cell morphometric measurements were conducted. On the 18th day, the presence of CTCs was verified using immunofluorescence with cytokeratin 20 (CK20) and plastin 3 (PLS3) markers. The results indicated that CTCs cultured in media supplemented with 10X ITS exhibited larger cell diameters compared to those cultured with 1X ITS and 10% FBS. This finding suggests that ITS plays a critical role in the successful culture of CTCs in serum-free media and that a 10X concentration of ITS can enhance the growth of colorectal cancer CTCs."

"Latar Belakang: Growth Differentiation Factor 9 (GDF9) adalah salah satu anggota dari superfamili TGF-β yang merupakan salah satu oocyte-secreted factor (OSF). GDF9 memainkan peran penting dalam folikulogenesis ovarium, kompetensi perkembangan oosit, serta sebagai molekul esensial yang mengontrol berbagai proses sel granulosa dan laju ovulasi. GDF9 merupakan OSF yang memainkan peran penting dalam menjaga fertilitas wanita. Penelitian ini bertujuan untuk mengetahui apakah ekspresi GDF9 berkorelasi dengan kualitas oosit dan laju fertilisasi pada pasien fertilisasi in vitro.
Tujuan: Mengetahui korelasi ekspresi GDF-9 dengan kualitas oosit dan laju fertilisasi pada pasien Fertilisasi In Vitro
Metode: Penelitian ini adalah penelitian potong lintang yang dilaksanakan di Klinik Yasmin, Rumah Sakit dr. Cipto Mangunkusumo, Jakarta sejak bulan Juli 2019 sampai bulan Juli 2020. Terdapat 26 wanita berusia 25-40 tahun pasien Fertilisasi In Vitro tanpa penyakit endometriosis maupun Sindrom Ovarium Polikistik yang menjadi subjek penelitian ini. Cairan folikel pasien diambil, kemudian dilakukan pengukuran ekspresi gen GDF9. Setelah itu, dilakukan evaluasi pada data ekspresi gen GDF9 serta diuji korelasinya dengan kualitas oosit dan laju fertilisasi pasien menggunakan aplikasi SPSS.
Hasil: Sebanyak 26 pasien IVF berpartisipasi dalam penelitian ini. Median jumlah ekspresi gen GDF9 adalah sebanyak 2.47 x 10-5 ng/μl dengan median kualitas oosit dan laju fertilisasi pasien IVF sebesar 3.00 dan 0.60. Berdasarkan hasil uji korelasi, terdapat korelasi negatif antara ekspresi GDF9 terhadap laju fertilisasi dengan kekuatan korelasi sedang (r = -0.443, p = 0.012). Sementara, ditemukan korelasi tidak bermakna antara ekspresi GDF9 terhadap kualitas oosit (r = -0.306, p = 0.064).
Kesimpulan: Terdapat korelasi negatif dengan kekuatan korelasi sedang antara ekspresi GDF9 dengan laju fertilisasi, sementara hubungan ekspresi GDF-9 dengan kualitas oosit memiliki korelasi tidak bermakna.

Background: Growth Differentiation Factor 9 (GDF9) is a member of the TGF-β superfamily which is one of the oocyte-secreted factor (OSF). GDF9 plays an important role in ovarian folliculogenesis, the competence of oocyte development, as well as an essential molecule that controls various granulosa cells process and the rate of ovulation. GDF9 is an OSF which plays an important role in maintaining female fertility. This study is conducted to see the correlation between the expression of GDF9 gene with the oocyte quality and the fertilization rate in the IVF patients.
Aim: To find out the correlation between the expression of GDF9 with the oocyte quality and the rate of fertilization of IVF Patients
Methods: This study is a cross-sectional study which was conducted at Klinik Yasmin dr. Cipto Mangunkusumo General Hospital, Jakarta since July 2019 to July 2020. 26 IVF patients aged 25-40 years without the conditions of endometriosis or Polycystic Ovary Syndrome (PCOS) participated in this study. Follicular fluids are taken from the patients and the GDF9 gene expression were measured. The data were then evaluated for its correlation with the oocyte quality and the fertilization rate using the SPSS application.
Results: Abour 26 IVF patients were participated in this study. The median amount of GDF9 gene expression was 2.47 x 10-5 ng/μl. The median of the oocyte quality and the fertilization rate was 3.00 and 0.60. According to the correlation test, there is a negative correlation between the expression of GDF9 gene and the fertilization rate with a moderate statistical correlation (r = -0.443, p = 0.012). Meanwhile, the expression of GDF9 gene has no significance correlation with the oocyte quality (r = -0.306, p = 0.064).
Conclusion: This study showed a negative correlation between the expression of GDF9 gene and the fertilizatio rate, while the relation between the GDF9 expression and the occyte quality has no significance correlation.

"Sel punca perinatal saat ini menjadi alternatif yang potensial bagi ketersediaan sel punca, baik dalam bidang riset maupun klinis. Salah satu yang diharapkan adalah Wharton's Jelly dari persalinan preterm yang merupakan sumber sel punca mesenkim. Karakteristik dan ekspresi antigen permukaan HLA-ABC dan HLA-G pada sel punca mesenkim Wharton's Jelly persalinan preterm belum banyak diketahui. Kedua molekul imunomodulator ini turut berperan dalam menentukan sifat supresi imun pada sel mesenkim, yang sangat dibutuhkan pada transplantasi allogenik. Penelitian ini bertujuan untuk mengetahui ekspresi antigen permukaan HLA-ABC dan HLA-G sel punca mesenkim WJ dari persalinan preterm dan dibandingkan pada berbagai suplemen medium kultur. SPM-WJ dikultur dalam medium DMEM 10 FBS, DMEM 10 PRP dan Mesencult. Sel yang telah konfluens dipanen, dan ditumbuhkan kembali pada wadah yang baru pasase dengan medium yang sama. Pada pasase 3 dan 5 dilakukan uji karakteristik antigen permukaan HLA-ABC dan HLA-G dengan menggunakan flowcytometry. Ekspresi HLA-ABC dan HLA-G sampel preterm Wharton's Jelly menunjukkan kecenderungan lebih hypoimmunogenic daripada sampel aterm dan lebih sesuai pada suplemen medium kultur PRP.Sel punca mesenkim WJ dari persalinan preterm dapat digunakan sebagai salah satu alternatif sumber sel punca mesenkim untuk aplikasi terapi regeneratif.

Perinatal stem cells is an potential alternative to the availability of stem cell, both in the research and clinical field. One would have expected is Wharton's Jelly from preterm delivery that is the source of mesenchymal stem cells. Characteristics and expression of surface antigen HLA ABC and HLA G on mesenchymal stem cells derived Wharton's Jelly from preterm delivery little has been known. Both of these immunomodulatory molecules play a role in determining the nature of immune suppression in mesenchymal stem cells, which are needed on the allogenic transplantation. This study aims to determine the expression of surface antigen HLA ABC and HLA G WJ mesenchymal stem cells from preterm delivery and compared on different culture media supplements. WJ MSC was cultured with the followings media DMEM 10 FBS, DMEM 10 PRP and Mesencult. Cells reaching confluence were harvested and regrown in different containers, but with the same media. Cell passaging was carried out until the fifth passage, and the characterization of HLA ABC and HLA G surface antigens were performed on the third and fifth passages. The expression of HLA ABC and HLA G at the Wharton's Jelly preterm samples tends more hypoimmunogenic than the term sample and more appropriate to the culture medium supplement PRP. Mesenchymal stem cells derived Wharton's Jelly from preterm delivery can be used as an alternative source of mesenchymal stem cells for regenerative therapy applications."

"ABSTRAK
Fungi Emericella nidulans strain MFW39 yang diisolasi dari ascidia Aplidium
longithorax dari Taman Nasional Laut Wakatobi, Sulawesi Tenggara memiliki aktivitas
sitotoksik terhadap beberapa sel lestari kanker. Senyawa yang berhasil diisolasi dan
bersifat sitotoksik adalah senyawa emestrin yang memiliki gugus ETP
(epipolithiodioxopiperazine). Senyawa emestrin memiliki beberapa jenis derivat. Pada
penelitian ini bertujuan untuk mengisolasi, mengidentifikasi dan menguji bioaktivitas
antikanker terhadap salah satu derivat senyawa emestrin. Dari hasil penelitian
didapatkan jenis derivat senyawa emestrin adalah emestrin B. Proses elusidasi struktur
dilakukan dengan metode HPLC, UPLC-MS, 1H-NMR dan 13C-NMR. Hasil uji
aktivitas sitotoksik dengan metode MTT (Microculture Tetrazolium Technique)
menunjukan IC50 fraksi emestrin B berkisar pada 0,18 􀂱 4,21 µg/mL. Urutan aktivitas
sitotoksik fraksi emestrin B terhadap sel lestari T47D [kanker payudara] (0,18 µg/mL) >
WiDr [kanker usus] (1,21 µg/mL) > HeLa [kanker serviks] (1,91 µg/mL). Fraksi
emestrin B juga bersifat sitotoksik terhadap sel lestari normal [Vero] (4,21 µg/mL). Oleh
karena itu fraksi emestrin yang berhasil diisolasi adalah emestrin B dengan rumus
struktur C27H22N2O10S3 dan memiliki potensi aktivitas antikanker.

ABSTRACT
Emericella nidulans marine fungi strain MFW39 was isolated from ascidia Aplidium
longithorax collected from Wakatobi Marine National Park, South East Sulawesi has a
biological activities to cancer cell lines. Emestrin was a compound with an ETP
(epipolithiodioxopiperazine) group that found in Emericella nidulans marine fungi have
cytotoxicity properties. Emestrin and another compound that related have the ETP
group. The research include isolation, identification and cytotoxicity assay of compound
that related with emestrin. Elucidation structure of molecule with HPLC, UPLC-MS,
1H-NMR and 13C-NMR. The fraction emestrin B have bioactivity to cancer cell lines in
range 0,18 􀂱 4,21 µg/mL with MTT (Microculture Tetrazolium Technique) assay method.
Order of anticancer activity was breast cancer cell line [T47D] (0,18 µg/mL) > colon
cancer cell line [WiDr] (1,21 µg/mL) > cancer cervic cell line [HeLa] (1,91 µg/mL). The
fraction of emestrin B have a toxicity to normal cell line [Vero] (4,21 µg/mL). The result
shows compound fraction that succeed to isolated was emestrin B (C27H22N2O10S3) and
have a potency anticancer activity."

"Latar belakang: xylitol merupakan gula alkohol (polyols) dengan 5 ikatan rantai karbon yang dilaporkan bermanfaat bagi kesehatan manusia. Dalam bidang kedokteran gigi, xylitol memiliki peran sebagai bahan antikaries gigi karena dapat menghambat pertumbuhan bakteri Streptococcus mutans. Namun, efek xylitol terhadap sel-sel pulpa gigi belum diketahui. Pulpa gigi merupakan jaringan yang sensitif terhadap paparan benda asing. Pada pulpa gigi yang terbuka, xylitol dapat
menimbulkan efek biologik sel.
Tujuan: untuk mendeteksi efek paparan xylitol terhadap protein total dan profil protein medium kultur sel-sel pulpa gigi.
Metode: sel-sel pulpa gigi didapat dari jaringan pulpa gigi sehat yang baru diekstraksi, kemudian dikultur dalam medium DMEM (37ºC, 5% CO2) hingga confluent. Kemudian dilakukan subkultur dengan kondisi yang sama selama semalam. Kelompok perlakuan dipaparkan xylitol dengan konsentrasi 2%, 4%, 8%, dan 16%, tetapi kelompok kontrol tidak dipapar xylitol. Konsentrasi protein total medium kultur sel-sel pulpa gigi diukur dengan menggunakan Bradford protein assay pada panjang gelombang 655 nm. Sedangkan profil protein medium kultur sel-sel pulpa gigi dianalisis dengan teknik SDS PAGE.
Hasil: rerata konsentrasi protein total (µg/ml ± SD) medium kultur sel-sel pulpa gigi pada kelompok perlakuan xylitol 2% (24.253,71 ± 2.363,29), xylitol 4% (21.925,42 ± 1.001,38), xylitol 8% (25.456,51 ± 4.569,45), dan xylitol 16% (26.306,66 ± 5.550,82) secara statistik dengan Oneway ANOVA lebih rendah bermakna (p<0,05) dibandingkan dengan kontrol (33.395,64 ± 4.209,08). Dari hasil SDS PAGE, ternyata tidak terjadi perubahan profil protein medium kultur sel-sel pulpa gigi setelah pemaparan xylitol.
Simpulan: konsentrasi protein total medium kultur sel-sel pulpa gigi menurun setelah pemaparan dengan xylitol, namun profil protein medium kultur sel-sel pulpa gigi tidak mengalami perubahan.

Background: xylitol is one of sugar alcohol (polyols) with 5 carbon atoms which is reported to have benefits to our health. In dentistry, xylitol has anti-caries effect as the growth of Streptococcus mutans could be inhibited. However, the xylitol effects on dental pulp have not been known yet. Dental pulp tissue is sensitive to foreign substances. Xylitol could penetrate the exposed dental pulp and induce the biological response of the cells.
Objective: to detect the effects of xylitol on dental pulp cells determined by total protein and protein profile of culture medium of the dental pulp cells (in vitro).
Methods: dental pulp cells were obtained from healthy and freshly extracted teeth. Then, they were cultured in DMEM medium (37ºC, 5% CO2) until confluent approximately 2 days. Subsequently they were subcultured and used as samples. The treatment groups were treated with xylitol 2%, 4%, 8%, and 16% and incubated at 37°C, 5% CO2 for overnight, while the control groups without xylitol. The total protein of culture medium was determined by Bradford protein assay in 655 nm. Whereas, the protein profile of culture medium were analized by SDS PAGE method.
Results: the mean of total protein? concentration (µg/ml ± SD) of culture medium in treatment groups with xylitol 2% (24.253,71 ± 2.363,29), xylitol 4% (21.925,42 ± 1.001,38), xylitol 8% (25.456,51 ± 4.569,45), dan xylitol 16% (26.306,66 ± 5.550,82) were lower than control group (33.395,64 ± 4.209,08). The comparison between the controls and treatment groups were analysed by Oneway ANOVA. All the treatment groups were signifcantly different compared with the controls (p<0,05). By SDS PAGE, the protein profile of culture medium in all treatment groups was not altered.
Conclusion: the total protein? concentration of culture medium of the dental pulp cells were decreased after treated with xylitol. However, the protein profile of culture medium of dental pulp cells was not altered.

"ABSTRAK
Makin berkurangnya energi fosil mendorong penelitian untuk mencari energi
alternatif. Sel fotovoltaik atau sel surya merupakan salah satu energi alternatif yang masih
dikembangkan. Sel surya menghasilkan energi listrik dengan mengubah energi dari cahaya
matahari. Dalam aplikasinya dibutuhkan pengertian yang mendalam mengenai karakter
sel surya sebelum dapat dimanfaatkan secara luas. Arus dan tegangan (I-V) yang dihasilkan
ketika sel surya diberikan cahaya intensitas dan pada suhu tertentu menjadi kajian dasar
utama untuk menentukan karakter sel surya. Besarnya intensitas cahaya dan suhu sel surya
harus diberikan sesuai standar umumnya penggunaan surya yaitu pada spektrum cahaya
matahari AM 1.5G dengan intensitas 1000 W/m2 dan suhu 25oC karena kondisi tersebut
mempengaruhi kinerjanya. Dalam sepuluh tahun terakhir pengembangan alat untuk
menguji karakter sel surya sangat banyak. Salah satunya dengan menggunakan MOSFET
untuk menaikkan karakter tegangan dan arus sel surya. Akan tetapi kebanyakan terutama
dipasaran masih mengunakan alat-alat yang mahal dalam membuatnya. Pada penelitian
tugas akhir ini ingin dibuat alat yang low cost untuk mengetahui karakter I-V (arus dan
tegangan) sel surya dengan metode MOSFET hanya dengan mikrokontroler AVR dengan
platform Arduino namun tidak mengurangi keandalan sistem dalam melakukan
pengukuran.

ABSTRACT
Decrasing of fuel energy lead to alternative energy research. Solar cell or photovoltaic
cell is one of many alternative energy which is still being developed. Solar cell works by
converting sunlight into electricity. In the application, it needs deep knowledge about solar
cell’s character before it can be any use. Current and voltage (I-V) which are produced
when the light hits the solar cell surface with a value of intensity and at a value of
temperature become the basic study to determine solar cell characters. The value of light
intensity and temperature must be given to the common standard of where and when solar
cell used. It is at AM 1.5G sunlight spectrum with 1000 W/m2 of intensity and temperature
at 25oC. In the past decade, there were so many development to make a device which
characterized solar cell. One of them uses MOSFET by raising electronic load to observe
solar cell current and voltage response. Yet many devices which has beed developed even
device on the market using many expensive tools. Hence in this thesis research, low cost
electronic controlled device for solar cell characterization is built with MOSFET method
and Arduino platform AVR microcontroler but still with high reliabity."
