Penyakit tuberkulosis yang juga dikenal dengan TB adalah penyakit paru-paru akibat kuman bakteri Mycobacterium tuberculosis. Bakteri Mycobacterium tuberculosis yang menginfeksi paru juga dapat menginfeksi bagian lain dari tubuh pasien, salah satunya adalah jaringan tulang belakang, yang disebut dengan penyakit TB tulang belakang. Salah satu obat anti tuberkulosis adalah Isoniazid (INH), yang mempunyai tantangan terbesar pada kepatuhan pasien meminum obat selama terus-menerus dalam suatu waktu tertentu. Pada penelitian ini, digunakan tiga variasi berbeda untuk menentukan formulasi hidrogel bermuatan INH, yaitu persentase asam malat, loading INH, dan jumlah siklus freeze-thaw, dan perhitungan dilakukan dengan optimasi Response-Surface Methodology (RSM). Didapatkan hasil bahwa rilis INH dirilis maksimal dengan formulasi asam malat 10%, loading OAT sebanyak 5%, dan siklus freeze-thaw sebanyak 4 kali siklus. Formulasi rilis terkecil didapatkan dengan asam malat sebanyak 6%, loading OAT sebanyak 15%, dan siklus freeze-thaw sebanyak 2 kali siklus. Formulasi rilis maksimal dapat dicapai hingga 67%, dan formulasi rilis minimal dapat dicapai sebesar 46%. Variabel yang paling signifikan diantara variabel lainnya adalah loading INH didalam matriks hidrogel. Matriks ini diharapkan dapat menjadi solusi rilis berkelanjutan bioimplant untuk penyakit TB tulang belakang.
Tuberculosis, also known as TB, is a lung disease caused by the bacterium Mycobacterium tuberculosis. Mycobacterium tuberculosis bacteria that infected the lungs can also infect other parts of the patient's body, one of which is spinal tissue, which is called spondilitis tuberculosis. One of the anti-tuberculosis drugs is Isoniazid (INH), which has the biggest challenge to the patient's compliance with taking medication continuously for a certain time. In this study, three different variations were used to determine the hydrogel formulation containing INH, namely the percentage of malic acid, INH loading, and the number of freeze-thaw cycles, where the calculation was done by Response-Surface Methodology (RSM). The results showed that the release of INH was released to the maximum with the formulation of malic acid 10%, OAT loading by 5%, and the freeze-thaw cycle by 4 times. The smallest release formulation was obtained with 6% malic acid, 15% OAT loading, and the freeze-thaw cycle by 2 times. The maximum release formulation can be achieved up to 67%, and the minimum release formulation can be achieved at 46%. The most significant variable among other variables is INH loading in the hydrogel matrix. This matrix is hoped to be a sustainable bioimplant release solution for spinal TB disease.
